Glutaredoxins (Grxs) are small ubiquitous oxidoreductase protein whose function in E.coli, yeast and mammals are well characterized but in plants still to be characterized. A cDNA band corresponding to Glutaredoxin was isolated by mRNA Differential Display PCR technique when Capsicum chinense plant was infected with Pepper Mild Mottle virus-Italian strain (PMMoV-I) virus. We examined the crucial function of Cch Grx gene against (PMMoV-I) virus and the resistance mechanism displayed by these gene in plants.
Confocal microscopy study showed that Cch Grx was targeted to chloroplast. Cch Grx induced resistant in N.benthamiana plants against the virus by the activation of SA-mediated defense-related genes (PR1, PR5 and PR2a). Expression of Cch Grx also altered the Pyridine Nucleotide contents towards reduced condition which enhances the resistance. Yeast transformation studies showed that Cch Grx is not found to be involved in the Fe-S cluster assembly. The protein was found to participate in deglutathionylation reaction found through in vitro assay. The gene was found to possess abiotic resistant property.
Cch Grx expression in N.benthamiana plants inhibits PMMoV-I viral accumulation and it was brought out by the induction of SA-regulated PR proteins and also by the alteration of Pyridine Nucleotide contents. These suggest that Cch Grx may alter the redox state of the TGA protein, a transcription factor involved in SA-mediated PR gene expression. The gene also able to rescue the abiotic stress conditions in plants but not able to participate in Fe-S cluster assembly of proteins.
